That isn't how it works, as the ground cable attaching the engine block to the battery would negate that circuit, not to mention the coolant temp sender wire, the oil pressure sender wire, etc. The polarity difference between opposite towers on the same coil is due to the secondary coil windings. Without delving too far into this, one side of the coil will output a positive voltage in relation to ground, while the other side will output a negative voltage in relation to ground. Disconnecting one plug wire will only disable the ignition of one cylinder.
